【摘要】:ThisEuropeanprestandardspecifiesamethodforthequantitativedeterminationofsaxitoxin,dc-saxitoxinandthequalitativedeterminationofneo-saxitoxin,andthegonyautoxinsGTX-2andGTX-3inmussels.ThemethodcanalsobeusedtoidentifythetoxinsC-1,C-2,GTX-5andGTX-6afterhydrolysisand,ifthesetoxinsarepresent,toexcludefalsepositiveresultsforGTX-2,GTX-3,neo-saxitoxinandsaxitoxin.Formusselthelowestlimitofdeterminationisforsaxitoxin0,04mg/kgmusselmeatandfordc-saxitoxin0,03mg/kgmusselmeat(signal/noise=10).Theupperlimitsofdeterminationhavenotbeendetermined.ThelimitsofdetectionforC-1,C-2,GTX-2,GTX-3,GTX-5,GTX-6andneo-saxitoxinhavenotbeendetermined.PSP(ParalyticShellfishPoisoning)toxinsareextractedfrommusselshomogenatewithanacidicaqueoussolution.Aftercentrifugationthesupernatantispurifiedbysolidphaseextraction(SPE)overaC18clean-upcartridge.PartoftheextractisdirectlyinjectedontheHPLCfortoxindetermination.Aftertheseparationontheanalyticalcolumn,postcolumnderivatisationisusedandthederivatizedtoxinsaredetectedusingfluorimetricdetection.HPLCanalysisisseparatedin2parts:asystemtoseparatethesaxitoxingrouptoxins(saxitoxin,dc-saxitoxinandneo-saxitoxin)andasystemtoseparatetheGTXgrouptoxins(GTX-2andGTX-3).IdentificationafterhydrolysisisbasedonthetransformationoftheN-sulfo-carbamoyltoxinstotheircorrespondingcarbamatetoxinsdetection.#,,#
